Tests thymosin β4 (Tβ4) for protecting brain microvascular endothelial cells from hypoxia-induced dysfunction using in vitro and TBI mouse models. Tβ4 stabilized blood-brain barrier (BBB) integrity under hypoxic conditions through an S1PR1 (sphingosine-1-phosphate receptor 1)-dependent mechanism—not previously described for Tβ4. Tβ4 treatment maintained tight junction proteins, reduced barrier permeability, and improved neurological outcomes after TBI. Identifies S1PR1 as the mechanistic link between Tβ4 and BBB protection in ischemic/traumatic brain injury.
