Intervertebral disc degeneration — a leading cause of back pain — is driven by inflammation and breakdown of the disc's structural proteins. A hydrogel-based delivery system was developed to release both an RNA inhibitor targeting a pro-inflammatory gene (miR-21) and the mitochondria-protective peptide SS-31 into degenerated discs in a sustained, controlled manner. In a rat model, this dual approach reduced inflammation, protected mitochondria from oxidative stress, and promoted regeneration of the disc's structural tissue.
