In rats subjected to chronic unpredictable stress — a model of depression — daily treatment with Semax or Melanotan II reversed key symptoms including loss of pleasure (anhedonia), abnormal weight changes, enlarged adrenal glands, and reduced levels of the brain protein BDNF, which is associated with mood and memory. Both peptides work through melanocortin receptors without triggering the hormonal effects of natural ACTH. The results support further exploration of this peptide class as a treatment for depression and stress-related disorders.
