RNA sequencing of rat brain tissue showed that both Semax and the related peptide ACTH(6-9)PGP primarily suppressed immune-related gene activity under normal (non-injury) conditions, with the two compounds having overlapping but distinct effects. Differences between the peptides were concentrated in genes tied to ion channel regulation and neural signaling. The study emphasizes the importance of understanding how these peptides affect the healthy brain, not just the injured brain, when evaluating their safety and specificity.
