Tests small spleen peptides (SSPs) for autoimmune disease tolerance, identifying thymosins (including thymosin β4) as primary SSP constituents. SSPs transformed dendritic cells from pro-inflammatory to tolerogenic through modulation of extracellular ATP/adenosine ratio. Recombinant thymosin peptides alone were less effective than the full SSP mixture—suggesting synergistic action among multiple thymosin family members. Provides mechanistic understanding of how thymosin peptides regulate DC differentiation and immune tolerance in autoimmune arthritis.
