In vitro study using human (LHCN-M2) and murine (C2C12) muscle progenitor cells to demonstrate that MOTS-c accelerates skeletal muscle differentiation into myotubes, with identification of a putative SH2 binding motif (YIFY region) in MOTS-c's sequence and use of a Y8F mutant to establish this region's role in myogenic differentiation signaling. First characterization of MOTS-c as a myogenic differentiation factor. Establishes that MOTS-c directly promotes muscle progenitor differentiation—providing a mechanism for MOTS-c's anti-sarcopenic effects that operates through regenerative (myogenic) rather than purely metabolic pathways, with implications for muscle regeneration after injury or in muscular dystrophies.
