Tests the effects of thymosin β4 silencing on biomechanical properties and stemness features of A375 melanoma cells. TB4-silenced melanoma cells showed altered cell stiffness (measured by atomic force microscopy) reflecting changes in actin cytoskeleton organization, but these biomechanical changes were not directly correlated with altered stemness markers (CD44, CD133, Sox2). Demonstrates that TB4's effects on cytoskeletal mechanics and cancer stemness are dissociable processes in melanoma—important for understanding TB4's multifaceted role in cancer.
