This review discusses the underestimated coding potential of mitochondrial DNA, highlighting the discovery of mitochondria-derived peptides like MOTS-c and humanin. These peptides, encoded by small open reading frames previously thought to be non-coding, challenge the conventional view that mitochondrial DNA only encodes 13 proteins and suggest additional functional genetic elements remain to be discovered.
