Identifies BRG1 (the ATPase subunit of the SWI/SNF chromatin remodeling complex) as required for Wt1 transcription factor expression in epicardial-derived progenitor cells (EPDCs) during cardiac development and regeneration. BRG1 and Wt1 were reactivated by thymosin β4 treatment and MI in adults, mediating EPDC differentiation into coronary smooth muscle. Reveals chromatin remodeling as the epigenetic mechanism by which TB4 and cardiac injury signals reactivate the fetal epicardial gene program—a fundamental mechanistic insight for TB4-driven epicardial cardiac regeneration.
