Uses 1-microsecond molecular dynamics simulation and antimicrobial assays to characterize fragment 9-19 of thymosin β4 as a scaffold for anti-biofilm agents. The fragment adopted hydrophobic core conformations suitable for membrane interaction. Optimized derivatives showed anti-biofilm activity against Staphylococcus spp. and Pseudomonas aeruginosa. Identifies TB4's central hydrophobic core peptide as an antimicrobial scaffold—opening a new therapeutic direction using TB4-derived fragments for treating biofilm-associated infections.
