Develops an intein-based biotechnological method to produce recombinant thymosin β4 with the required N-terminal acetylation in E. coli. The system uses an in vivo N-acetyltransferase combined with intein self-splicing for protein cleavage and purification, producing rhTβ4 at lower cost than chemical synthesis. Provides an alternative intein-based approach to the separate acetyltransferase expression system (PMID 29989423)—both addressing the key manufacturing challenge of producing biologically active N-acetylated TB4 at pharmaceutical scale.
