Describes the discovery and characterization of CJC-1295 as a long-lasting GHRH analog that works by covalently binding to the free thiol group on albumin after injection. Three maleimido derivatives of human GRF(1-29) were synthesized; CJC-1295 emerged as the most stable and potent, with enhanced resistance to enzymatic degradation and prolonged GH-stimulating activity in rats.
