This study demonstrated that thymosin alpha-1 significantly enhances human sperm fertilizing capacity. T-alpha-1 increased sperm penetration rates in the sperm penetration assay by promoting capacitation and acrosome reaction. Antibodies to thymosin alpha-1 bound primarily to the acrosomal region of sperm and also enhanced penetration rates. Both thymosin alpha-1 and beta-4 were detected in seminal plasma of fertile men, with thymosin alpha-1 levels being significantly lower in infertile men with defective sperm function, suggesting potential diagnostic and therapeutic applications for male infertility.
