This study demonstrated that serum from tumor-bearing mice contains factors that induce thymocyte apoptosis in a time- and dose-dependent manner, involving FasL and TNFα signaling. The apoptotic pathway involved protein kinases and phosphatases, with increased pro-apoptotic proteins (p53, Bax, Bad, Fas) and Bcl-2 cleavage. Importantly, thymosin alpha-1 was shown for the first time to antagonize this tumor-induced thymocyte apoptosis by activating protein kinase C, decreasing pro-apoptotic Bax and Bad levels, and increasing Bcl-2 expression.
