Develops a collagen-chitosan sponge scaffold (CCSS) with encapsulated thymosin β4 for controlled delivery in diabetic wound healing with hindlimb ischemia. The TB4-loaded scaffold was fabricated by freeze-drying, characterized by electron microscopy, and tested in streptozotocin-diabetic rats with combined ischemia and excisional wounds. CCSS-eTβ4 significantly accelerated wound closure and angiogenesis versus scaffold alone or systemic TB4. Demonstrates scaffold-based local delivery as superior to systemic administration for diabetic wound healing—a clinically meaningful advance for the challenging combined ischemia/hyperglycemia wound environment.
